چکیده

diopathic dilated cardiomypathy (DCM) is characterized y the progressive depression of cardiac function and left entricular dilation in the absence of coronary artery disease. here is evidence to suggest that humoral immunity plays n important role in the pathogenesis of DCM. In particlar, the role of autoantibodies in the pathogenesis of DCM as been emphasized (1). Sera from patients with DCM how a variety of autoantibodies, including those against embrane proteins, mitochondrial proteins, heat-shock roteins, and myocyte structural sarcolemmal proteins. Beause all these antibodies are not present in each patient ith DCM, it is apparent that all these autoantibodies are ot pathogenic in DCM. To date, only 2 autoantibodies ave satisfied the criteria of cause-and-effect relationship for CM, and both these antibodies display stimulatory activties. The first comes from a report that program death eceptor-1 (PD-1) mice develop autoantibodies against ardiac troponin 1 (cTpn 1) and dilated heart, and second rom the finding that the injection of autoantibodies against Tpn-1 from these mice into normal mice induces dilation nd dysfunction of the heart in recipient mice (2), therefore stablishing a direct role of anti–cTpn-1 antibody in dilaion and dysfunction of heart resembling human DCM.

Fc receptors and their interactions with immunoglobulins.

Receptors for the Fc domain of immunoglobulins play an important role in immune defense. There are two well-defined functional classes of mammalian receptors. One class of receptors transports immunoglobulins across epithelial tissues to their main sites of action. This class includes the neonatal Fc receptor (FcRn), which transports immunoglobulin G (IgG), and the polymeric immunoglobulin rece...

Role of Fc Receptors as a therapeutic target.

It has been forty years since the discovery of Fc Receptors and their function. Fc Receptors include the IgG receptors (FcgammaR), high-affinity IgE receptor (FcepsilonRI), IgA and IgA/IgM receptors, and neonatal Fc receptor for IgG (FcRn). In particular, the FcgammaRs have been well known to play an important role in many biologic processes including those associated with the response to infec...

Activating and inhibitory Fcγ receptors in rheumatoid arthritis: from treatment to targeted therapies

Fc gamma receptors (Fc gammaRs) bind the constant Fc region of IgG molecules. IgG/antigen-containing immune complexes elicit a variety of effector functions in cells that express activating Fc gammaRs. Because activating Fc gammaRs are present on cells from the innate immune system, such as dendritic cells, monocytes/macrophages and granulocytes, these IgG receptors form a crucial link between ...

Regulation of high-affinity IgE receptor-mediated mast cell activation by murine low-affinity IgG receptors.

Allergic symptoms result from the release of granular and lipidic mediators and of cytokines by inflammatory cells. The whole process is initiated by the aggregation of mast cell and basophil high-affinity IgE receptors (Fc epsilon RI) by IgE and antigen. We report here that IgE-induced release of mediator and cytokine can be inhibited by cross-linking Fc epsilon RI to low-affinity IgG receptor...

Glycosylation and Fc receptors.

Immunoglobulins and Fc receptors are critical glycoprotein components of the immune system. Fc receptors bind the Fc (effector) region of antibody molecules and communicate information within the innate and adaptive immune systems. Glycosylation of antibodies, particularly in the Fc region of IgG, has been extensively studied in health and disease. The N-glycans in the identical heavy chains ha...
